DoNotAge Longevity Sachets Review: What’s Inside?

I started trying out the DoNotAge sachets back in May 2026.  I've been taking loads of different longevity supplements for a while now.  Originally I started taking NAD+ precursors like NMN and NR when I was going through IVF treatment, and now continue to take them for health reasons.  I really liked the idea of having everything in one sachet rather than having to take dozens of different capsules every day.

The DoNotAge longevity sachets contain 15 ingredients.  You mix it up in water and drink one a day.  I have a short video that I posted on YouTube showing what comes in the box and how to make the drink.  In this review I'll tell you about my experience and also what each sachet contains.

I want to point out that I have a thyroid condition and I was concerned that the sachet contains Quercetin and Resveratrol in quite high doses, which are both thyroid disruptors. For that reason I've been monitoring my thyroid blood tests since I started taking the sachet.

I also want to say at the start that I do have a partnership with DoNotAge but I am always honest in all my reviews and show exactly which supplements I take and won't take.  You can see my review of the DoNotAge individual supplements here.

Key Takeaways

  1. DoNotAge longevity sachets contain 15 ingredients that target all 12 hallmarks of aging, including genomic instability, cellular senescence, and mitochondrial dysfunction.
  2. The sachets contain 1,000mg of NMN, which is a higher amount than comparable products like IM8 Longevity (600mg) and TMRW (500mg).
  3. Quercetin and resveratrol in the sachets may act as thyroid disruptors when taken in high doses, which is a consideration for individuals with thyroid conditions.
  4. DoNotAge includes SIRT6Activator from Fucus vesiculosus seaweed, which is currently the only proven activator of sirtuin 6 protein linked to DNA repair.
  5. The sachets include high molecular weight hyaluronic acid based on research showing naked mole rats produce hyaluronic acid molecules over five times larger than humans, potentially contributing to their 30-year lifespan and cancer resistance.

What are DoNotAge longevity sachets?

DoNotAge is a UK-based longevity supplement company selling individual ingredients and a combined daily formula, that comes in a sachet. The sachets include 15 longevity compounds that target all the 12 hallmarks of aging.

In each subscription cycle you get enough sachets to last four weeks. In the box is a shaker, a storage box called the Box For Life and a routine tracker stamp.

The main attraction for me is having fewer things to organise. You can look through one ingredient list and decide whether the combination suits you.

Glycine

Glycine helps the body make collagen and glutathione, an antioxidant that protects cells. It has extended lifespan in mice and supports skin elasticity.

Sirt 6 Activator

SIRT6 is a protein studied in relation to DNA repair, cellular integrity and inflammatory pathways. DoNotAge’s SIRT6Activator comes from Fucus vesiculosus, a seaweed compound source. It is the only proven activator of sirtuin 6.

NMN

DoNotAge verifies every batch of its pure NMN at 99.8% purity. On the DoNotAge website you can see the certificate of analysis showing that it is the purest NMN.

NMN is a direct precursor to nicotinamide adenine dinucleotide, usually shortened to NAD. This coenzyme participates in cellular energy metabolism and the activity of enzymes involved in DNA repair.

TMG

TMG supports methylation, a process involved in gene regulation.

Why TMG and NMN Work Better Together

DoNotAge includes both 1,000mg NMN and 1,000mg TMG in equal amounts, and this pairing isn't random as it addresses a critical biochemical challenge that most people taking NAD+ precursors don't know about.

The Hidden Cost of NAD+ Boosting:
When your body converts NMN into NAD+, it uses methylation to process and recycle the nicotinamide byproduct. This process consumes methyl groups—essential chemical units your body needs for:
• DNA methylation and gene regulation
• Neurotransmitter production
• Detoxification pathways
• Homocysteine metabolism

The Methylation Drain Problem:
Taking high-dose NMN without methylation support can potentially deplete your methyl donor pool, especially with doses of 1,000mg daily. This is where people might experience:
• Elevated homocysteine levels
• Reduced methylation capacity for other critical processes
• Diminished benefits from NAD+ supplementation over time

How TMG Solves This:
TMG (trimethylglycine) provides three methyl groups per molecule, directly supporting the methylation cycles that NMN metabolism demands. The 1:1 ratio in DoNotAge sachets ensures you're not creating a biochemical bottleneck while boosting NAD+.

Timing Consideration:
Both compounds are water-soluble and work synergistically when taken together. The sachet format ensures you're never taking one without the other, which maintains methylation balance throughout your supplementation period.

Why Most Companies Skip This:
TMG adds cost and requires larger serving sizes. Many NMN-only products leave this methylation support gap, expecting users to address it separately—if they even know about it.

Ca-AKG

Ca-AKG helps cellular metabolism and plays a role in releasing energy from food.

Trans-resveratrol

Resvetrarol activates Sirt 1, an enzyme found mostly in the cell nucleus that regulates metabolism, gene expression, and stress responses. You can buy a pure trans-resveratrol supplement separately on the DoNotAge website.

Nitralis

Nitralis is designed to boost nitric oxide, which helps blood vessels relax and supports cardiovascular health. If you take blood pressure medication, check with your doctor before taking it.

Quercetin

Pure Quercetin may help clear damaged cells that linger, and provide antioxidant and anti-inflammatory benefits.

Magnesium bisglycinate

Magnesium bisglycinate provides magnesium, which supports energy production, normal muscle and nerve function, and healthy bones. Supports brain health.

SulforaBoost

SulforaBoost is broccoli seed extract and increases sulforaphane levels in the body. This targets the oxidative stress pathways as it switches on the body's own antioxidant defences. It is also used to support detoxification.

hyaluronic acid

Hyaluronic acid is often found in skincare products, but you can also take it as a supplement for cellular longevity, joint integrity and skin health.

It was research involving naked mole rats that brought attention to the role of hyaluronic acid in longevity.

Naked mole rats produce hyaluronic acid molecules over five times larger than those in humans or mice. They also break HA down more slowly, allowing it to build up in their tissues.

When researchers removed these molecules from cells in the lab, the cells became more vulnerable to cancerous changes. This suggests the molecules help explain the animals’ unusual cancer resistance.

Naked mole rats can also live for over 30 years, which is unusually long for a small rodent. It is believed that their unusually large hyaluronic acid molecules may contribute to that longevity by protecting tissues.

Check out the Ted Talk below about the clinical trials into hyaluronic acid.

Hyaluronic Acid Molecular Weight Guide

Molecular WeightPrimary FunctionBest For
Low (5-50 kDa)Penetrates tissues, wound healingSkin hydration, joint mobility
Medium (50-300 kDa)Immune modulation, inflammation controlBalanced cellular effects
High (1,000-2,000 kDa)Cellular protection, anti-inflammatoryLongevity, cancer resistance
Very High (2,000+ kDa)Structural support, mechanical protectionTissue integrity, space filling
Naked Mole Rat (6,000+ kDa)Cancer resistance, exceptional longevityResearch model only

Why Molecule Size Matters More Than Amount

Most supplement companies focus on dosage amounts, but DoNotAge took a different approach with their hyaluronic acid inclusion and they focused on molecular weight instead. This decision stems from groundbreaking naked mole rat research that revealed a fascinating longevity mechanism.

What Makes Naked Mole Rat HA Special:
• Produce HA molecules 5x larger than human molecules
• Break down HA significantly slower than other mammals
• Live 30+ years (10x longer than similar-sized rodents)
• Show remarkable cancer resistance linked to large HA molecules

The Science Behind Size:
When researchers removed large HA molecules from naked mole rat cells in laboratory settings, those cells became vulnerable to cancerous changes. This suggests the protective effect isn't just about having hyaluronic acid—it's about having the right molecular structure.

Why This Changes Supplementation Strategy:
Standard hyaluronic acid supplements often contain low molecular weight HA (under 50 kDa) because it's cheaper and easier to absorb. DoNotAge specifically uses high molecular weight HA (typically 1,000-2,000 kDa) to more closely mimic the protective molecules found in long-lived species. This distinction matters because different molecular weights interact with different cellular receptors and trigger distinct biological responses.

The Trade-Off:
High molecular weight HA may have lower bioavailability, but emerging research suggests that even partial absorption or gut-level effects may provide systemic benefits through immune modulation and cellular signaling pathways.

CoQ10

Coenzyme q 10 helps mitochondria produce energy and acts as an antioxidant, helping protect cells from oxidative damage. Supports heart health.

Vitamin K2

Vitamin K2 supports healthy bones by activating proteins involved in calcium regulation. It also activates a protein that helps limit calcium build-up in blood vessels.

Vitamin D3

Vitamin D supports bone health and contributes to the normal function of the immune system.

Spermidine

Spermidine is linked to autophagy which is the process cells use to clear out and recycle damaged components. It is important for cellular protection.

The Sirtuin Spectrum Strategy: Activating Different Longevity Pathways Simultaneously

DoNotAge includes both SIRT6Activator and trans-resveratrol in the same formula, which might seem redundant since both are "sirtuin activators." However, this combination reveals a sophisticated understanding of how different sirtuins work in different cellular locations.

The Sirtuin Family Breakdown:

SIRT1 (Activated by Resveratrol):
• Location: Primarily cell nucleus
• Function: Gene expression, metabolism, stress response
• Best known for: Mimicking caloric restriction effects

SIRT6 (Activated by Fucus vesiculosus):
• Location: Cell nucleus, chromatin
• Function: DNA repair, telomere maintenance, inflammation control
• Unique feature: Only proven direct SIRT6 activator available

Why Activate Both?
SIRT1 and SIRT6 don't compete - they complement each other by working on different aspects of cellular aging:
• SIRT1 focuses on metabolic switching and energy management
• SIRT6 concentrates on genomic stability and cellular integrity
• Together they address both energy metabolism AND structural cellular maintenance

The Fucus Vesiculosus Advantage:
Most supplements focus exclusively on SIRT1 activation through resveratrol or other polyphenols. DoNotAge's inclusion of seaweed-derived SIRT6Activator is significant because:
• SIRT6 specifically protects against genomic instability (a primary aging hallmark)
• SIRT6 activity naturally declines with age more dramatically than other sirtuins
• No other commercially available compound reliably activates SIRT6

Dosage Context:
At 500mg trans-resveratrol and 1,600mg SIRT6Activator, DoNotAge provides clinical-range doses for both pathways. Most longevity-focused individuals would need to purchase these as separate supplements, making the sachet format particularly efficient for multi-sirtuin activation protocols.

The Research Gap:
While resveratrol has decades of research, SIRT6 activation is an emerging area with most studies still in animal models. The combination represents both established science (SIRT1) and cutting-edge research (SIRT6) in one formula.

The 12 hallmarks of ageing

Ageing involves more than getting wrinkles or feeling tired. There are 12 different changes that happen inside our cells that contribute to how we age. These include DNA damage, declining energy production and less efficient cellular clean-up.

DoNotAge sachets contain ingredients that target each of these 12 hallmarks.

HallmarkWhat it meansIngredient to target this hallmark
Genomic instabilityDNA damage builds upSIRT6Activator, NMN
Telomere attritionProtective chromosome ends shortenSIRT6Activator, resveratrol
Epigenetic alterationsChanges in how genes are switched on and offSIRT6Activator, Ca-AKG
Loss of proteostasisProtein maintenance becomes less efficientSulforaBoost, glycine
Disabled macroautophagyCells struggle to recycle damaged componentsSpermidine
Deregulated nutrient sensingCells respond differently to nutrientsCa-AKG, TMG
Mitochondrial dysfunctionCellular energy production declinesCoQ10, NMN

Research into healthy aging mechanisms is early, and much of it is in cells or animals rather than the human body. Human studies show NMN raises NAD in the blood, which supports healthy NAD levels and cellular health. Evidence that it protects cognitive function or slows neurological decline is limited.

My Experience taking the DoNotAge Sachets

I have several blood tests a year to check my thryoid hormones and after I started the sachets I noticed that my thyroid hormone levels were the worst they have been in ages. I have hypothyroidism so this probably won't affect most people but I wanted to mention it. I'm currently having a break for four months and will then have another blood test to see if the levels are the same.

I also previously stopped taking Nitralis as it was causing me to have dizzy spells and I noticed Nitalis is in the sachet. I had a few dizzy spells while taking the sachet so I wondered if that was why. Most people don't seem to have this side effect though.

Overall I did notice improved energy levels and it was much more convenient being able to take the sachet instead of dozens of supplements. I also took the sachets with me to Italy and it was really useful to not have to pack loads of different supplement bottles.

Common Side Effects and Mitigation Strategies

Potential Side EffectLikely IngredientWho Should Monitor
Dizziness, light-headednessNitralis (nitric oxide booster)Those on blood pressure medication
Thyroid hormone changesQuercetin, resveratrol (high doses)Hypothyroid or hyperthyroid individuals
Digestive discomfortHigh glycine or magnesiumSensitive digestion, IBS

Sleep changesNMN timing (circadian impact)Take in morning, not evening

How easy are they to use?

The sachet is easy to use if you dislike swalling multiple capsules. They are also really convenient when you're travelling. I found I had to stir it a lot to mix the powder fully with the water and some of it doesn't completely dissolve. It tastes like berries and I personally didn't have a problem with the taste. Two ingredients that are missing are creatine monohydrate and collagen peptides, but you can mix these in separately.

FAQ

Can I take the DoNotAge sachets if I'm already taking individual NMN or resveratrol supplements?

You should avoid doubling up on the same ingredients, as the sachets already contain clinical doses of NMN (1,000mg) and resveratrol (500mg). Taking additional amounts could push you into excessive doses without added benefit. If you want to continue individual supplements, you may need to alternate days or choose products that complement rather than duplicate the sachet ingredients.

Why doesn't the powder completely dissolve in water?

Some ingredients in the sachets, particularly fat-soluble compounds like CoQ10 and resveratrol, don't fully dissolve in water regardless of how much you stir. This is normal and doesn't affect the efficacy - you're still consuming the ingredients, just in suspension rather than solution. Some users add a small amount of healthy fat (like MCT oil) to help with the fat-soluble component absorption, though this isn't required.

Is there an optimal time of day to take the sachet for maximum benefit?

Morning consumption is generally recommended because NMN may influence circadian rhythm and energy metabolism, potentially interfering with sleep if taken late in the day. The sachets also contain ingredients that support daytime functions like nitric oxide production and cellular energy. Taking it with or after breakfast can help with absorption of fat-soluble ingredients and reduce any potential digestive sensitivity.

How long does it take to notice any changes after starting the sachets?

Effects vary significantly by individual and which markers you're monitoring. Some people report increased energy within 1-2 weeks, while structural changes like improved skin quality may take 8-12 weeks. Since the sachets target cellular-level aging processes, many benefits aren't immediately perceptible and would only show up in blood work, biological age tests, or long-term health outcomes. I monitored thyroid levels specifically due to pre-existing conditions, which revealed changes within months.

Can the sachets replace a multivitamin, or should I still take both?

The sachets contain only vitamins D3 and K2, so they don't replace a comprehensive multivitamin that includes B vitamins, vitamin C, vitamin E, and minerals like zinc and selenium. The formula focuses on longevity-specific compounds rather than basic nutritional coverage. If your diet doesn't provide adequate micronutrients, you should continue a separate multivitamin or ensure you're getting these nutrients from whole foods.

Important Insights to Remember

  1. Thyroid function requires monitoring when taking high-dose quercetin and resveratrol as both compounds can act as thyroid disruptors, particularly for individuals with pre-existing hypothyroidism or hyperthyroidism who should check thyroid blood markers regularly.
  2. Molecular weight matters more than dosage amount for hyaluronic acid supplementation because naked mole rat research demonstrates that larger HA molecules (over 5x human size) provide superior cancer resistance and longevity benefits compared to standard low molecular weight forms.
  3. NMN requires methylation support through TMG to prevent biochemical depletion since converting 1,000mg of NMN daily consumes significant methyl groups needed for DNA regulation, neurotransmitter production, and homocysteine metabolism.
  4. SIRT6Activator from Fucus vesiculosus represents the only commercially available compound that directly activates SIRT6 protein which specifically addresses genomic instability and telomere maintenance differently than SIRT1 activation from resveratrol.
  5. The sachet format delivers clinical-range doses across 15 ingredients targeting all 12 hallmarks of aging which would otherwise require purchasing and organizing separate bottles costing £120-180 monthly versus the consolidated £80-100 sachet subscription.
